YAP1(Yes-associated protein 1)是一种转录共激活因子,属于Hippo信号通路的核心效应分子,同时也是Hippo-YAP信号通路的关键调控基因。该基因家族属于TAZ/YAP家族(原英文:TAZ/YAP family),其成员包括YAP1和TAZ(WWTR1),两者在功能上高度相似,均通过结合TEAD转录因子家族(原英文:TEAD family)调控下游靶基因的表达,参与细胞增殖、分化、凋亡及组织再生等生物学过程。YAP1的生物学功能主要体现在促进细胞生长、抑制凋亡、维持干细胞特性及参与器官大小调控。其表达产物YAP1蛋白在细胞质与细胞核之间穿梭,当Hippo通路失活时,YAP1进入细胞核,与TEAD结合激活靶基因(如CTGF、CYR61等),促进细胞增殖;反之,当Hippo通路激活时,YAP1被磷酸化滞留于胞质并降解。YAP1的主要作用位点包括上皮组织、肝脏、心脏和骨骼肌等,尤其在胚胎发育和组织修复中发挥关键作用。突变对YAP1功能的影响显著,例如功能获得性突变(原英文:gain-of-function mutation)可导致YAP1持续激活,引发细胞过度增殖,与多种癌症(如肝癌、乳腺癌、结直肠癌)密切相关;而功能缺失性突变(原英文:loss-of-function mutation)则可能导致发育缺陷或组织再生障碍。YAP1过表达会驱动肿瘤发生、转移及耐药性,同时抑制细胞凋亡,其机制涉及对细胞周期调控基因(如cyclin D1)的上调;而降低表达(如通过siRNA或药物抑制)可抑制肿瘤生长,但可能影响正常组织的再生能力。YAP1与Hippo通路其他成员(如MST1/2、LATS1/2)形成调控网络,其异常表达还会影响Wnt、TGF-β等信号通路的交叉作用。在疾病方面,YAP1的异常激活与纤维化疾病、心血管疾病及神经退行性疾病也有关联。该基因家族的共性是依赖TEAD转录因子调控靶基因,并响应机械力、细胞极性等物理化学信号,协调组织稳态与再生。
