SRC基因是一种原癌基因(proto-oncogene),属于SRC家族酪氨酸激酶基因家族(SRC family kinases,SFKs),该家族成员(如LYN、FYN等)均编码非受体型酪氨酸激酶(non-receptor tyrosine kinases),通过磷酸化靶蛋白调控细胞信号传导。SRC基因编码的SRC蛋白在细胞增殖、分化、黏附、迁移等过程中起核心作用,其活性受C端酪氨酸残基(Tyr530)磷酸化的负调控,当该位点去磷酸化时,SRC激酶被激活。SRC突变(如Tyr530突变或激酶结构域突变)会导致组成性激活(持续活化),促进肿瘤发生,与结肠癌、乳腺癌等多种癌症相关。SRC过表达会增强下游信号通路(如MAPK、PI3K/AKT),导致细胞异常增殖和转移;而敲低SRC表达会抑制肿瘤生长,但可能影响正常组织的伤口愈合和骨代谢(因SRC参与破骨细胞活化)。SRC家族基因的共性是含有SH3、SH2结构域(介导蛋白相互作用)和激酶结构域(催化磷酸化),并通过调控细胞骨架重组和整合素信号影响肿瘤侵袭性。目前针对SRC的靶向药物(如达沙替尼)用于治疗白血病,但需注意其可能引起的心脏毒性等副作用。
This gene is highly similar to the v-src gene of Rous sarcoma virus. This proto-oncogene may play a role in the regulation of embryonic development and cell growth. The protein encoded by this gene is a tyrosine-protein kinase whose activity can be inhibited by phosphorylation by c-SRC kinase. Mutations in this gene could be involved in the malignant progression of colon cancer. Two transcript variants encoding the same protein have been found for this gene. [provided by RefSeq, Jul 2008]
