MAP4K3(丝裂原活化蛋白激酶激酶激酶激酶3)属于STE20丝氨酸/苏氨酸蛋白激酶家族,是MAPK信号通路的上游调节因子。该基因编码的蛋白通过激活下游的MAPK级联反应参与多种细胞过程,包括细胞增殖、分化、凋亡和应激反应。MAP4K3在mTORC1信号通路中发挥关键作用,通过感知氨基酸水平调节细胞生长和代谢。其主要作用位点包括细胞质和细胞膜,通过与Rag GTPases相互作用调控溶酶体上的mTORC1活性。MAP4K3突变可能导致其激酶活性改变,影响mTOR信号传导,与癌症、代谢性疾病和神经退行性疾病相关。研究发现MAP4K3过表达会异常激活mTOR通路,促进肿瘤细胞生长和存活,而敲低MAP4K3则抑制mTOR活性,导致细胞生长受阻。在2型糖尿病中,MAP4K3过表达会加剧胰岛素抵抗。MAP4K3属于GCK(生发中心激酶)亚家族,该家族成员均含有高度保守的激酶结构域,通过磷酸化下游底物调控应激反应和细胞周期。这个家族的特点是通过级联放大信号参与多种生理病理过程。MAP4K3还与自噬调控密切相关,其表达水平变化会影响细胞清除受损蛋白质和细胞器的能力。在神经系统中,MAP4K3参与调节神经元存活和突触可塑性,其异常表达可能与阿尔茨海默病等神经退行性疾病有关。
