HNF1B(肝细胞核因子1β,英文全称Hepatocyte Nuclear Factor 1 Beta)是一种转录因子基因,属于HNF1基因家族,该家族还包括HNF1A。HNF1家族基因的共同特点是编码的蛋白质含有同源异型结构域(homeodomain,一种能结合DNA的蛋白质结构)和二聚化结构域,主要调控与代谢、发育相关的基因表达。HNF1B在胚胎发育和成人组织中发挥重要作用,尤其在肾脏、胰腺、肝脏和生殖系统的发育和功能维持中至关重要。HNF1B的表达产物通过与特定DNA序列结合,调控下游靶基因的转录,影响细胞分化、器官形成和葡萄糖代谢等过程。HNF1B的主要作用位点包括肾脏的肾小管上皮细胞、胰腺的β细胞以及肝胆管细胞。HNF1B突变会导致其功能异常,与多种疾病相关,最常见的是青少年发病的成人型糖尿病5型(MODY5),这是一种常染色体显性遗传病,表现为早发性糖尿病和肾脏异常如肾囊肿。此外,HNF1B突变还与先天性肾脏和尿路畸形(CAKUT)、多囊肾、胰腺发育不全以及生殖系统异常如子宫畸形或精子发生障碍有关。HNF1B过表达可能干扰正常发育信号通路,导致组织过度增生或功能紊乱,例如在肾脏中可能促进囊肿形成;而降低表达则可能引起器官发育不全或功能障碍,如胰腺β细胞减少导致胰岛素分泌不足。HNF1B的表达变化还会影响其他基因,例如下调GLUT2(葡萄糖转运蛋白2)和胰岛素基因的表达,进一步扰乱血糖调控。在癌症中,HNF1B的异常表达与某些卵巢透明细胞癌和肾细胞癌相关,可能通过影响细胞增殖或代谢途径参与肿瘤发生。由于HNF1B的多效性,其突变或表达失调对机体的影响复杂,涉及多个器官系统,临床表现为高度异质性。

This gene encodes a member of the homeodomain-containing superfamily of transcription factors. The protein binds to DNA as either a homodimer, or a heterodimer with the related protein hepatocyte nuclear factor 1-alpha. The gene has been shown to function in nephron development, and regulates development of the embryonic pancreas. Mutations in this gene result in renal cysts and diabetes syndrome and noninsulin-dependent diabetes mellitus, and expression of this gene is altered in some types of cancer. Multiple transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Sep 2009]
