CD247基因编码T细胞受体(TCR)复合物的ζ链(CD3ζ),是T细胞表面TCR-CD3复合体的关键组成部分。TCR-CD3复合体负责识别抗原并激活T细胞免疫应答。CD247的生物学功能主要体现在它与CD3γ、δ、ε链共同形成CD3复合物,将TCR与抗原结合的信号传递到细胞内,启动下游信号通路(如ZAP-70激酶活化),最终导致T细胞增殖、分化及细胞因子释放。其主要作用位点是T细胞膜,与TCRαβ或TCRγδ异二聚体非共价结合。CD247突变会导致严重免疫缺陷,如T细胞信号传导障碍,表现为反复感染或自身免疫病。它与某些疾病密切相关,例如CD247表达异常与系统性红斑狼疮(SLE)等自身免疫疾病相关,其低表达可能导致免疫耐受失衡。若CD247过表达,可能增强T细胞活性,但过度激活会引发自身免疫反应;若表达降低,则导致T细胞功能缺陷,增加感染风险或肿瘤免疫逃逸。CD247属于CD3基因家族,该家族成员(CD3γ、δ、ε、ζ)均编码TCR-CD3复合物的跨膜蛋白,共同特点是胞内段含有免疫受体酪氨酸激活基序(ITAM),用于传递抗原识别信号。CD3家族基因的共性是通过ITAM结构域募集下游信号分子,调控T细胞发育和功能。专业术语解释:TCR(T细胞受体)是T细胞识别抗原的膜蛋白;ITAM(免疫受体酪氨酸激活基序)是信号转导中磷酸化的酪氨酸残基模体;ZAP-70是一种参与T细胞激活的酪氨酸激酶。若术语"ζ链"翻译不准确,可备注为CD3ζ chain。

The protein encoded by this gene is T-cell receptor zeta, which together with T-cell receptor alpha/beta and gamma/delta heterodimers, and with CD3-gamma, -delta and -epsilon, forms the T-cell receptor-CD3 complex. The zeta chain plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways. Low expression of the antigen results in impaired immune response. Two alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
