BRMS1(乳腺癌转移抑制基因1,Breast cancer metastasis suppressor 1)是一种肿瘤转移抑制基因,最初在乳腺癌中被发现,其表达产物通过调控多种信号通路抑制肿瘤细胞的转移能力。BRMS1的生物学功能主要包括通过影响核因子κB(NF-κB,一种调控炎症和细胞存活的转录因子)的活性来抑制肿瘤细胞的侵袭和转移,同时还能通过恢复缝隙连接(gap junctions,细胞间通讯结构)的功能促进细胞间信号传递,从而抑制肿瘤进展。BRMS1主要作用位点在细胞核和细胞质,通过与其他蛋白质相互作用发挥功能。当BRMS1发生突变或表达降低时,其抑制肿瘤转移的功能减弱,可能导致肿瘤细胞更具侵袭性,与乳腺癌、卵巢癌、肺癌等多种癌症的转移和不良预后相关。相反,过表达BRMS1可显著抑制肿瘤细胞的转移潜能,但对正常细胞影响较小。BRMS1属于一个功能相关的肿瘤转移抑制基因家族,这些基因的共性是通过不同机制阻断肿瘤转移的多个步骤,如细胞迁移、侵袭和定植。BRMS1还通过表观遗传调控(如组蛋白去乙酰化,histone deacetylation,一种影响基因表达的修饰方式)影响下游基因的表达。此外,BRMS1的表达受微小RNA(miRNA)等调控分子的影响,其表达水平变化可能进一步影响肿瘤微环境(tumor microenvironment,肿瘤周围的细胞和分子网络)及其他相关基因的功能。研究显示,BRMS1在多种癌症中表达下调,恢复其表达可能成为抑制肿瘤转移的治疗策略。

This gene reduces the metastatic potential, but not the tumorogenicity, of human breast cancer and melanoma cell lines. The protein encoded by this gene localizes primarily to the nucleus and is a component of the mSin3a family of histone deacetylase complexes (HDAC). The protein contains two coiled-coil motifs and several imperfect leucine zipper motifs. Alternative splicing results in two transcript variants encoding different isoforms.
